Homicide Squad detectives have launched an investigation, with the exact circumstances surrounding the incident yet to be determined.

No arrests have been made at this stage.

A crime scene has been established and Bourke Street remains closed between Elizabeth Street and Queen Street.

In a separate incident around 300m away, two men were hospitalised after a wild brawl broke out outside the Inflation nightclub earlier on Sunday morning.

Police said the men were found injured about 1.25am following a large affray at the intersection of Flinders Lane and King Street, with the Herald Sun reporting the fight is estimated to have involved more than 100 people.

A 19-year-old male with suspected serious injuries and an unidentified man with life-threatening injuries have both been taken to hospital.

A crime scene has been set up and the area cordoned off while detectives examine the scene, with officers from the Major Crime Scene Unit also set to attend.

Photos taken at the scene show blood smears on a car parked outside the nightclub.

Police are investigating whether the two stabbings just hours apart are linked.

In a third earlier incident, three men were left injured after an assault in Southbank.

Police have been told two groups of men appeared to argue as they were leaving a bar along Riverside Quay about 12.40am.

The argument allegedly spilled onto the promenade footpath, where three men were set upon by two other men.

A 25-year-old man from Springvale was struck heavily to the face before he fell and hit his head on the ground.

The two other men, aged 24 and 25-years from Keysborough and Cheltenham, received facial injuries.

The pair who police said appeared to be the aggressors fled the scene.

Anyone who witnessed any of the incidents, has video footage or further information is urged to contact Crime Stoppers on 1800 333 000.
